Assuming you are filling it on a normally hot day 80-100 degrees outside, then the the pressure will not vary widely to where you will not get an accurate fill.
In terms of system efficiency, assuming you are running the AC in the summer, then the it's mostly effected by the ability to cool the condenser. This is dependent on the water vapor in the air, as hot, humid air has a higher enthalpy than dry air. If you are not using recirculate then heat transfer from the evaporator would be affected as well
