Sort of. If everything is ideal, that is the case, but if something isn't or the car is experiencing temperature issues, the three runs won't be the same. Three gives you some help diagnosing any issues that may crop up that wouldn't normally.
Really though, that's just a side benefit. Real reason is that the labor to strap the car down and do one run is the same as for three, so might as well do three and make sure your car is running well and making consistent power.
Last time I got dynoed was in 105F temp with no humidity at altitude. First run was about 5 hp up compared to third run, back to back, even with fan blowing at the car, so there is some definite variability inherent just from atmospheric conditions.
